Well, you've run into the problem where there are people that say stuff like that in all seriousness, so just assuming it would come off as sarcasm doesn't work.
There are plenty of people that really would seriously say something as stupid as people thinking that just because the star of a movie is not white, that he couldn't possibly speak English. (We'll just ignore all the Jackie Chan and Jet Li movies these people would have been exposed to.) Fact is that we've had a number of movies in the last handful of years that were either originally in another language or originally were. (Just off the top of my head, Pan's Labyrinth and Passion of the Christ.) And since in the previews of Ninja Assassin, if I recall, all you see are Asian people.
Some people would seriously assume that racism absolutely MUST be the answer for any questions arising about the film's origin and language (generally because some people assume that racism must be the answer to any question), when the more likely explanation is just that they're wanting to know if it originated in another country and has subtitles, like, say, Hero.
